چکیده

The contributions of Stamey in the area of prostate cancer diagnosis, monitoring, and treatment and in the prostatespecific antigen (PSA) literature are numerous and noteworthy.’ His accompanying Opinion article [1] deals with aspects of analytical performance as well as clinical interpretations of PSA assays and raises concern that clinicians may be misled by recently published data. We agree with many of his concerns and disagree with others. In areas of disagreement, we would like to offer alternative views that are based on extensive published and unpublished data from our own research group.

Trends in immunoassays of prostate-specific antigen: serum complexes and ultrasensitivity.

Multiple reviews and 1200 papers have appeared in the literature describing prostate-specific antigen (PSA).’ Without question, this has become the most prominent tumor marker in oncology. Since the introduction of the first commercial PSA immunoassays 7 years ago, the antigen has been an invaluable marker for monitoring patients with prostate cancer. Furthermore, during the past few years, det...

Ultrasensitive assay of prostate-specific antigen used for early detection of prostate cancer relapse and estimation of tumor-doubling time after radical prostatectomy.

We used an ultrasensitive prostate-specific antigen (PSA) assay with a detection limit of 0.02 microgram/L for long-term monitoring of PSA changes in 5 patients who were cured by radical prostatectomy and in 10 patients who had failed prostatectomies; 5 patients who underwent cystoprostatectomy were also evaluated with one sample after surgery. Relapse-free periods, determined on the basis of c...

Preparation of prostate specific antigen standards for immunoradiometric assay

Background: Immunoradiometric assay is one of the most common and precise methods for determination of prostate specific antigen (PSA) in clinical laboratories. Usual use of human serum in routine assays has many disadvantages such as easy contamination and precipitation, instability and unavailability. Thus in order to avoid these problems the artificial matrix was used which acts sim...

Age Specific Reference Levels of Serum Prostate-Specific Antigen, Prostate Volume and Prostate Specific Antigen Density in Healthy Iranian Men

Background: It is relevant to highlight that there is not a precise and perfect report on either 95 percentile value (upper limit of normal range) or on appropriate reference intervals for serum PSA in Iranian population. Objective: To determine age-specific reference ranges for serum prostate-specific antigen (PSA) concentration and PSA density (PSAD) and prostate volumes in a population of he...

Ultrasensitive detection of prostate-specific antigen by a time-resolved immunofluorometric assay and the Immulite immunochemiluminescent third-generation assay: potential applications in prostate and breast cancers.

We report an ultrasensitive time-resolved immunofluorometric assay (TRIFA) for prostate-specific antigen (PSA). The assay is an improvement of our previous report (Clin Chem 1993;39:2108-14) and includes the utilization of two monoclonal antibodies and a one-step incubation period, which greatly reduces analysis time. The new method demonstrates a superior lower analytical limit of detection (<...
